چکیده

Introduction ELISPOT assays are primarily used to detect the number of T cells that respond to a given antigen. With that number being absolute for any given donor sample, ELISPOT counts should be similar between laboratories, if subjectivity in counting is avoided. Due to the differences in spot sizes ranging from microns to millimeters, setting cut offs for minimal and maximal spot sizes will lead to substantial variability between investigators when determined subjectively, irrespective of experience. In contrast, if spot size distributions would follow predictable statistical functions, objective gating decisions could be made using common standards, eliminating subjective calls of the counting process. This study aims to determine if ELISPOT size distribution follows predictable statistical distributions and therefore if ELISPOT counting can be made objective based on statistical principles.

High Reproducibility of ELISPOT Counts from Nine Different Laboratories

The primary goal of immune monitoring with ELISPOT is to measure the number of T cells, specific for any antigen, accurately and reproducibly between different laboratories. In ELISPOT assays, antigen-specific T cells secrete cytokines, forming spots of different sizes on a membrane with variable background intensities. Due to the subjective nature of judging maximal and minimal spot sizes, dif...

User vs. software-dependent variability of ELISPOT counts obtained from ten different laboratories

Introduction In each human donor’s PBMC, there is a defined number of T cells specific for any given antigen. A major goal of immune monitoring with ELISPOT is to measure this number accurately and reproducibly between different laboratories. In ELISPOT assays, cytokine spots produced by antigen-specific T cells show a broad spectrum of sizes and densities over variable background. Therefore, e...

ELISPOTs Produced by CD8 and CD4 Cells Follow Log Normal Size Distribution Permitting Objective Counting

Each positive well in ELISPOT assays contains spots of variable sizes that can range from tens of micrometers up to a millimeter in diameter. Therefore, when it comes to counting these spots the decision on setting the lower and the upper spot size thresholds to discriminate between non-specific background noise, spots produced by individual T cells, and spots formed by T cell clusters is criti...

ELISPOTs produced by CD8 and CD4 cells follow log normal size distribution

Introduction Each positive well in ELISPOT assays always contain spots of variable sizes that range from micrometers up to a millimeter in diameter. Therefore, when it comes to counting ELISPOTs it is critical to decide how to set the lower and the upper spot size thresholds to discriminate between non-specific background noise, spots produced by individual T cells, and spots formed by T cell c...

Log-Normal and Mono-Sized Particles’ Packing into a Bounded Region

Many systems can be modeled with hard and various size spheres, therefore packing and geometrical structures of such sets are of great importance. In this paper, rigid spherical particles distributed in different sizes are randomly packed in confined spaces, using a parallel algorithm. Mersenne Twister algorithm was used to generate pseudorandom numbers for initial coordination of particles. Di...
